Consequently, the following results were obtained: 1. Monoclonal antibody (MoAb) MUSE11 reacted with the synthetic peptide corresponding to the tandem repeat domain of human polymorphic epithelial mucin (PEM). 2. A cDNA clone of 735bp was isolated from a normal human kidney cDNA library and was designated as mucin 700 by using an oligonucleotide probe corresponding to part of the tandem repeat domain. Nucleotide sequencing showed that mucin 700 consisted of 10 tandemly repeated domains. 3. Recombinant mucin 700 was produced with an expression vector, pGEX-2T, in E. Coli. MoAb MUSE11 was shown to react with this protein by Western blot analysis and sandwich enzyme immunoassay. 4. PEM core protein mRNA was expressed in both cancerous and non-cancerous regions of the stomach and the colon. It was revealed by Northern blot analysis using total RNAs extracted from surgically resected tissues.
