The Src family kinases, p56lck (Lck) and p59fyn (Fyn), were expressed in insect cells from baculovirus vectors. The peptides that contain sequences found at the tyrosine-based activa-tion motifs of the CD3-ζ chain and the Ig-α chain, a PDGF-receptor peptide, and angiotensin II were in vitro labeled with the kinases added as an immune complex with anti-Lck and anti-Fyn conjugated to Sepharose beads. The Lck and Fyn kinases expressed in insect cells were autophosphorylated on tyrosine and found to be fully active. In the immune complex kinase assay, more than 40% of the radioactivity added as [y-??P]ATP were incorporated into these pe-ptides, which were isolated by HPLC after the reaction.
