We aimed here to detect cancer-related gene mutations in routine fecal specimen; dried feces spots (DFS) on a filter paper especially designated for an automated fecal occult blood (FOB) test. DNA was simply extracted from a 3 mm disc of DFS (including around 10 mg feces) with a boiling procedure followed by a cationic surfactant extraction. K-ras common mutations, as the typical cancer-related gene alterations, were detected with a nested allele specific PCR with microfluorometry (ASP/MFL). Adequate DNA was recovered from 363/451 (80.5%) DFS specimens tested. The K-ras mutations were detected in 2/24 (8.3%) from patients with colorectal cancer and 13/315 (4.1%) from screening participants with positive FOB. No mutations were detected in 24 DFS specimens with negative FOB. The results of K-ras mutation frequency were not encouraging to achieve practically a specific dreliable method for colorectal cancer screening. However this report, identifying for the first time a cancer related gene alteration in feces using identical samples with routine FOB test, will be a step to realize a molecular genetic testing for the cancer screening.","subitem_description_type":"Abstract"}]},"item_7_identifier_registration":{"attribute_name":"ID登録","attribute_value_mlt":[{"subitem_identifier_reg_text":"10.15114/tr.36.7","subitem_identifier_reg_type":"JaLC"}]},"item_7_publisher_32":{"attribute_name":"出版者","attribute_value_mlt":[{"subitem_publisher":"Sapporo Medical University"}]},"item_7_source_id_7":{"attribute_name":"ISSN","attribute_value_mlt":[{"subitem_source_identifier":"0041-4093","subitem_source_identifier_type":"ISSN"}]},"item_7_version_type_15":{"attribute_name":"著者版フラグ","attribute_value_mlt":[{"subitem_version_resource":"http://purl.org/coar/version/c_970fb48d4fbd8a85","subitem_version_type":"VoR"}]},"item_creator":{"attribute_name":"著者","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{"creatorName":"Yamaguchi, Akihiro"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Hashimoto, Naoko"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Tsutae, Wataru"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Seino, Kuniyoshi"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Osanai, Hiroyuki"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Yamamoto, Naoya"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Tokino, Takashi"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Sato, Noriyuki"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Kikuchi, Kokichi"}],"nameIdentifiers":[{}]}]},"item_files":{"attribute_name":"ファイル情報","attribute_type":"file","attribute_value_mlt":[{"accessrole":"open_date","date":[{"dateType":"Available","dateValue":"2019-07-31"}],"displaytype":"detail","filename":"n004140933617.pdf","filesize":[{"value":"93.1 kB"}],"format":"application/pdf","licensetype":"license_note","mimetype":"application/pdf","url":{"label":"n004140933617.pdf","url":"https://sapmed.repo.nii.ac.jp/record/14190/files/n004140933617.pdf"},"version_id":"68926916-42b7-4189-a894-413f548e39a0"}]},"item_keyword":{"attribute_name":"キーワード","attribute_value_mlt":[{"subitem_subject":"Stool","subitem_subject_language":"en","subitem_subject_scheme":"Other"},{"subitem_subject":"Noninvasive sample","subitem_subject_language":"en","subitem_subject_scheme":"Other"},{"subitem_subject":"Fecal occult blood","subitem_subject_language":"en","subitem_subject_scheme":"Other"},{"subitem_subject":"Colorectal cancer","subitem_subject_language":"en","subitem_subject_scheme":"Other"},{"subitem_subject":"K-ras mutation","subitem_subject_language":"en","subitem_subject_scheme":"Other"},{"subitem_subject":"Genetic screening ","subitem_subject_language":"en","subitem_subject_scheme":"Other"}]},"item_language":{"attribute_name":"言語","attribute_value_mlt":[{"subitem_language":"eng"}]},"item_resource_type":{"attribute_name":"資源タイプ","attribute_value_mlt":[{"resourcetype":"departmental bulletin paper","resourceuri":"http://purl.org/coar/resource_type/c_6501"}]},"item_title":"Dried Feces Spots as an Alternative DNA Source : Detection of K-ras Mutations in Colorectal Cancer Screening","item_titles":{"attribute_name":"タイトル","attribute_value_mlt":[{"subitem_title":"Dried Feces Spots as an Alternative DNA Source : Detection of K-ras Mutations in Colorectal Cancer Screening","subitem_title_language":"en"}]},"item_type_id":"7","owner":"15","path":["1663"],"pubdate":{"attribute_name":"PubDate","attribute_value":"2019-07-31"},"publish_date":"2019-07-31","publish_status":"0","recid":"14190","relation_version_is_last":true,"title":["Dried Feces Spots as an Alternative DNA Source : Detection of K-ras Mutations in Colorectal Cancer Screening"],"weko_creator_id":"15","weko_shared_id":-1},"updated":"2023-12-13T02:43:37.121254+00:00"}
