This antibody was shown to react immunohistochemically with adenocarcinomas of the lung, the stomach and the pancreas. Among non-cancerous tissues, this antibody reacted very weakly with the renal tubulus and the pancreas only. On the other hand, the antigen corresponding to this antibody was observed as being widely distributed in the fetal tissues using the immunoperoxidase technique. Periodic acid treatment on the tissue section completely decreased the reactvity of this antibody, suggesting that the antibody might recognize the carbohydrate determinant. Neuraminidase digestion of the tissue section unmasked the determinant in some portions of the cancerous parts which were negatively stained before neuraminidase digestion. These results suggest that the nature of the carbohydrate epitope is \"cryptic\". Molecular size of the antigen detected in the surpernatant of A549 cells was estimated to be over 330K daltons by using sodium dodecyl sulfate polyacrylamide gel electrophoresis and western blotting. Gel filtraton analysis, employing enzyme-linked immunosorbent assay, also suggested the very high molecular weight of the antigen in the supernatant. Measurement of the antigen in the sera of cancer patients was performed by reversed passive hemagglutination assay. Only three of 30(10.0%) healthy donors and four of 31(12.9%) patients with benign diseases showed serum antigen levels of more than 1/64 dilution. In contrast, the positive frequency of the antigen (more than 1/64 dilution) was 45.0% to 73.3% in the sera of patients with lung and gastrointestinal cancers.
